Abstract Soluble MHC molecules are present in healthy human serum and body fluids in various concentrations which however change in pathological conditions. These molecules have been associated with the process of tolerance and with immunological regulation in general. In the present study sMHC-II molecules have been isolated from the supernatants of CD4+ cells. These cells came from either control or tolerized with HSA BALB/c mice and their role as to the regulation of the immune response was evaluated in vivo as well as in vitro. The sMHC-II molecules that were injected in immunized with HSA mice resulted in reduction of the anti- HSA antibody while they had no significant influence on the T cell specific markers. The sMHC-II molecules that were provided to HSA-immunized spleen cells resulted in reduced cell proliferation and downregulation of CD28, the marker of T-cell activation. They also induced an increase of the anti- HSA antibody in contrast with the in vivo experiments. The results presented here concentrate on three major conclusions: CD4+ cells secret sMHC-II molecules, sMHC-II repress the humoral response in vivo while downregulating T-cells in vitro.
