Abstract This study focuses on the precipitation, separation and identification of proteins in wines and grape musts in Greek and foreign grape species, at different wine-making stages and in different areas. Specifically, proteins were isolated from aromatic white wines must as well as pink and red musts varieties. The species tested originate from different areas and were collected from different wine-making stages. However, they have under gone similar wine-making process. In order to precipitate proteins, various techniques were performed including precipitation with ammonium sulfate, polyethylene glycol (PEG) and trichloroacetic acid. After this stage, proteins were separated through one-dimensional and two-dimensional electrophoresis of polyacrylamide gel. Matrix assisted laser desorption / ionization (MALDI TOF/TOF) was used for this purpose. The aim of the study was their identification in order to find which proteins cause protein haze and is resistant to it. Electrophoresis helped to the protein determination separation of grape must and wine proteins first in different zones and to their identification afterwards. The majority of the proteins were identified in the area of small and medium molecular weight which seems to be responsible for the problem occuring in white wine musts. Proteins with molecular weight of approximately 24kDa and 32kDa were identified. Thaumatin and chitinase are two common proteins, that are abundant inextracts of mature grapes, leaves and roots. Finally, a protein with molecular weight of 71kDa was identified as vacuolar invertase.
