Abstract Blm protein is one of the major players in maintaining DNA integrity, something that is easily understood by the impact of its loss. Patients with the BLM-/- phenotype (Blooms Syndrome) are predisposed for all types of cancer encountered in the general population and their cells present strong genetic instability and loss of heterozygosity . Chk1 protein is an important regulator of cell cycle which activates in the event of DNA damage. Apart from this, it has an important role in orchestrating DNA repair by activating or by placing proteins during damage through its kinase activity. These two proteins have been found to interact in vivo, with Chk1 phosphorylating Blm at Ser646, localizing it to the PML bodies at the damage point during replication stress. In vitro, Chk1 can phosphorylate Blm at Ser502, which leads it to degradation. The functional role of this phosphorylation is however not known . The purpose of this study is to investigate the biochemical interaction between these two proteins, and finding the conditions that lead to the Blm degradation.
