Abstract Molecular diagnostics are a very useful tool for the management of insecticide resistance in mosquitoes. The aim of this study is the application of already known assays, and the development of new methods for monitoring the resistance and other traits of the mosquitoes vectors. Allele-specific PCR methods were applied to a greek population of Culex pipiens (main vector of West Nile Virus) in order to test the presence of the kdr mutation, L1014F (confers resitance to pyrethroids) and iAChE mutation, (G119S that confers resistance to carbamates and organophosphates). The frequency of kdr mutant alleles was high (50% of the total number of alleles) and the frequency of the iAChE mutation was low (8,3% of the total number of alleles) in the population checked. Additionally, a novel multiplex molecular assay was developed, for the simultaneous detection of several traits of Anopheles gambiae complex species (main vectors of malaria). The method was designed for the identification of (i) the species of An. gambiae complex, (ii) the molecular form (M or S) of An. gambiae s.s., (iii) the presence of kdr, iAChE and Rdl mutations and (iv) the presence of Plasmodium infection. Future work includes further development and optimization of the method described above. Finally, the molecular mechanism of resistance to temephos, using a resistant greek strain of Aedes albopictus (vector of many arboviruses) was examined.
