Abstract Atherosclerosis is a fibroproliferative disease of the artery intima. Several lines of evidence suggest that among other factors, genetic and molecular alterations are involved in the development of atherosclerosis. Initial studies have shown alterations in the DNA of atherosclerotic plaques, at the microsatellite level. In the present study, we extend our initial findings, performing a microsatellite analysis in 27 atherosclerostic plaques, using 25 highly polymorphic markers focused on chromosomes 1, 2, 8, 9 and 17. To our knowledge, this is the first microsatellite analysis using atherosclerotic plaques obtained from cerebral vessels. Our analysis revealed that specific loci on chromosomes 2, 8, 9 and 17 exhibited significant incidence of LOH. 46% of the specimens showed loss of heterozygosity (LOH) at 2p13-p21, 48% exhibited LOH at 8p12-q11.2 while at 8p11.2-p21.1 LOH was detected in 47% of the cases. Considerable incidence of LOH was observed at 17q21, 9q31-34, and 17p13, 39%, 31% and 27%, respectively. Interestingly, genetic alterations were detected in a more significant rate as compared to the corresponding alterations observed in plaques from other vessels. Our results indicate an elevated mutational rate, suggesting a potential implication of these chromosomal regions in atherogenesis
