Abstract This dissertation presents the development and application of new analytic techniques, based on spectroscopy NMR, for the detection of phenolic compounds in virgin olive oil. The first analytical technique involves the use of one-dimensional (1D) 1H-NMR spectra and two-dimensional (2D) H,H gCOSY NMR spectroscopy for the analysis of the polar part of virgin olive oil. The analysis of the spectra is based on the comparison with spectral parameters of model phenolic compounds, which have been obtained in the present dissertation. The analytical technique of 1H-NMR spectroscopy refers as well on the quality and the quantity of the phenolic compounds and it can identify a total of 16 phenolic compounds. The second analytical technique applied is based on the coupling of HPLC with NMR spectroscopy through a modified solid phase extraction system (LC-SPE-NMR). The application of this method confirmed the presence of 18 polyphenolic compounds in virgin olive oil, while the identification of 14 more new phenolic compounds has been achieved. The third analytical technique for the qualitative and quantitative determination of polyphenolic compounds in virgin olive oil is based on the phosphitylation of the hydroxyl groups of the phenolic compounds with a suitable reagent and the detection of the phosphytilated derivatives by employing 31P-NMR spectroscopy. By the application of this technique a great number of polyphenolic compounds has been identified and quantified in virgin olive oil.
