Abstract Recent studies have shown that inside the IMS of mitochondrion takes place an oxidative folding protein import pathway. In contrast to the other protein import pathways that depend on membrane potential and hydrolysis of ATP, this new import pathway depends on oxidative folding. The main role in this pathway has an oxidase of ΙMS, called Mia40. The oxidized Μia40 interacts with the precursor substrates and induces the intramolecular disulfides bonds and folding of the substrate. Then Mia40 gets reduced and has to be reoxidized. This reaction is carried out by Erv1. From the other hand Erv1 is also a substrate of Mia40. In this work our goal is to study the biogenesis of the Erv1 into the IMS. We saw that the C-terminal part of the Erv1 is responsible for the binding to Mia40, whereas the N-terminal has an ancillary role for the import process. Therefore the combinatorial function of these parts is necessary for the translocation across the outer membrane. In addition, we identified the part that interacts non-covalently with Mia40, using pepscan binding assay.
