Abstract |
The infection of plants by pathogens can have serious effects on growth, especially in the event that contamination carried by the early seedling stage . The interaction of the virus and the plant includes the potential interactions of virus (RNA or proteins) with host proteins and RNA of the virus with the host RNA . Viroids are models for studying the interaction between a non-coding genome and the host genome. Although it is small (250 - 400nt) and known structure, only in some cases it has been determined the exact mechanism of pathogenesis. During viroid infection, the root of the plants has been analyzed in terms of few studies symptoms , changes in gene expression and distinct stages of replication. Based on all these studied the multiplication of potato spindle tuber viroid (PSTVd) in their host Solanum lycopersicum ( or lycopersicum esculentum) cv Rentita ( tomato ) . The purpose was to clarify the differences in proliferation (quantity and quality ) of the virus-like leaves and root system . Plants infected with the strain of the viroid PSTVd KF440.2. Five experiments were conducted in a sand soil (3:1), two in culture perlite and two experiments in vitro cultivation of aseptic seed. In these cultures, followed by the processes of homogenization and extraction , to isolate the RNA material. Over this through methods of electrophoresis , the gel denaturing agarose - formaldehyde , 5% polyacrylamide and polyacrylamide 15% urea , separated circular to linear viroid and tried to observe possible occurrence of siRNAs. Finally to compare strains of the virus-like radical leaves conducted isolation for sequencing. The conclusions show the course of infection, the amount of viroid , a comparison of the viroid RNA forms between the leaf and the root, and the phenotypic difference appeared in intense symptoms in the aerial parts , in contrast to the basement remained approximately unchanged . Interestingly expected sequencing of RNA than the leaf and root.
