Abstract |
RNA silencing known also as RNA interference (RNAi) is an evolutionary conserved mechanism in virtually all eukaryotes and is implicated in sequence-specific regulation of gene expression as well as antiviral defence in higher plants and insects. Small RNAs (sRNAs, 21-24nt-long), including microRNAs (miRNAs) and small – interfering RNAs (siRNAs) are key components of RNAi mechanism acting as signals for complementarity – mediated sequence degradation. Hence, sRNAs are involved in the control of various plant developmental and defense processes.
In plants, SERRATE (SE) encodes a C2H2 zinc finger protein and is considered a post-transcriptional regulator of miRNAs levels. Experimental data indicate that direct interaction of SE with HYL and DCL1 promote accurate processing of pri-miRNA precursors in the plant cell nucleus. serrate mutants revealed a pleiotropic phenotype, affecting leaf polarity, abnormalities in phase transition, meristem activity and inflorescense architecture.
Here, we present the phenotypic analysis of SERRATE overexpressors and knockdown N.benthamiana and N.tabacum plants along with the response of these plants to viral and viroid infection. Knockdown SERRATE tobacco plants show abnormal leaf development and phase transition delay from vegetative to reproductive phase compared to wild type plants. Our experimental data also suggest that SERRATE and as a consequence miRNAs, participate in plant viral defense. Further studies are required in order to understand the pathway through which SERRATE affects plant defense.
