Abstract |
Coxiella burnetii is an obligate intracellular bacterium that causes Q fever in humans and
other mammals. Q fever is a worldwide zoonotic disease and can cause acute or chronic life-
threatening disease in humans. Acute Q fever is mostly a mild flu-like illness. In a percentage
of patients infected with C. burnetii, the primary infection will be developed into chronic Q
fever. Chronic Q fever leads to high mortality rates if left untreated, making preventive
measures critical for high-risk patients. Q fever has become a serious public health problem
in many areas, not previously known as endemic areas and poses a significant risk to human
and animal health. Therefore, the reliable diagnosis of the disease is essential. In the present
study, a number of C. burnetii antigenic proteins were tested for their diagnostic ability in a
number of patient sera. Five proteins have been selected that have been described
literature-specific for acute Q fever. The plasmids carrying the expression genes of these
proteins were constructed and transformed into E. coli cells. The expression conditions for
each protein, temperature, inducer concentration and time after induction were then
checked, and the optimal ones were determined. Isolation of the proteins was followed, and
their antigenicity was studied with serums of patients from acute and chronic Q fever, as
well as healthy blood donors. The sensitivity and specificity of these tests were determined
by statistical analysis. Based on these results, a protein was selected for immobilization on a
gold electrode and studied by electrochemical impedance spectroscopy. This device could
later be used as a serological diagnostic tool.
