Abstract |
Plasmodium berghei parasite has been extensively used for studying the physiology, life cycle and gene function of the malaria transmitting parasites. Adapting the well-characterized CRISPR/Cas9 editing system to that parasite provides an opportunity to elucidate the role of genes, revealing little by little information that may lead to a new drug or vaccine discovery for the elimination of malaria, a disease raging to endemic African regions for centuries. The newly characterized Oocyst Rupture Proteins of Plasmodium, and their ability to inhibit the rupture of the oocyst upon their absence, shown in knock-out experiments, reveal their potential as drug targets. In order to study more extensively their role and function, we exploit the system CRISPR/Cas9 gene editing in Plasmodium. Here, we attempted to generate two transgenic parasitic lines for studying ORP1 and ORP3 proteins in vivo. In addition, expression of the ORP proteins in bacteria, is a first step to reveal their molecular structure as a step to understand their function. C-CAP protein is yet another essential protein for the completeness of the life cycle of the Plasmodium in an earlier stage, a conditional knock out created with RGR CRISPR/Cas9 method may eventually reveal when and why is this protein needed. Lastly, a drug study of 60 plant-based Essential Oils, have revealed some chemical compounds that may act with an anti-parasitic role, leading to the discovery of a new drug against malaria.
