Abstract The transmission of mitochondrial DNA (mtDNA) in the vast majority of species is maternal. One theory explains that the advantage of maternal inheritance is due to the elimination of spreading of selfish mutations in the population. Despite the various mechanisms of inhibiting the paternal leakage of mtDNA , the phenomena of leakage and mitochondrial heteroplasmy are increasingly reported in the literature. Based on the prevailing hypothesis, the state of heteroplasmy enables recombination between different mtDNAs. The recombination of mtDNA may have significant evolutionary consequences, since very low-frequency recombination would be sufficient to reverse the effects of the continuous accumulation of deleterious mutations due to random genetic deflection (Muller's Ratchet). In the present study we investigated whether the phenomena of paternal leakage of mtDNA and the maternal inheritance of heteroplasmy are random processes or are significantly influenced by genetic factors. In order to answer our questions, we conducted a series of interspecific crosses in two generations of Drosophila genus. Our results show that the leakage of paternal mtDNA is significantly associated with factors such as sex, family, age and heteroplasmic state of the mother. We also observed that the maternal inheritance of heteroplasmy is significantly affected by the family. We conclude that differences in levels of parental leakage and maternal inheritance of heteroplasmy between individuals and families indicate that these phenomena are affected by specific, unknown genetic mechanisms.
