Abstract During aging, a multitude of changes occurs, directly affecting immune system integrity and creating an environment of low-grade, chronic inflammation irrespective of pathogen encounter. DNA damage plays a pivotal role in the process of cellular aging, triggering a cascade of stress responses that are sensed within the cell. Through intercellular communication, a series of phenotypic changes then emerge at a systemic level. Previous work in our lab by Goulielmaki et al. (2020) showed that DNA-repair deficient macrophages (in the Ercc1F/-; Lys-Cre+ mouse model) initiate a systemic pro-inflammatory response and metabolic reprogramming, which is accompanied by the secretion of extracellular vesicles (EVs). In this study, we focus on the communication of macrophages and CD4+ T cells, which together coordinate a productive immune response that aged individuals lack. First, we characterize the functionality of DNA-repair proficient CD4+ T cells in an altered inflammation environment. We find that T cells become hypo-functional in response to the accumulation of DNA damage in macrophages. Next, we suggest macrophages in this setting are both potential activators and, later, suppressors of CD4+ T cells, and we propose mechanisms by which this communication might be achieved.
