Abstract |
Polymerase chain reaction (PCR) is an enzymatic method of amplifying specific parts of genetic material in vitro. Speed, specificity, high sensitivity and low cost are the reason that PCR is established as one of the most commonly used methods both in research and diagnostic level. What is requested from PCR is high speed in reproduction, as well as fidelity in the samples created. Within this research area, Minotehbiotechnology- FORTH, developed a chimeric DNA polymerase called Minopol.
The aim of the present Thesis was to find the best working conditions of this polymerase, as well as the identification of its characteristics during a PCR reaction.
Minopol polymerase was tested in different conditions (5 different buffers), testing each time its performance. After that, there was an efficiency test of the polymerase in different DNA template concentrations (viral, plasmid and bacterial DNA), and also identification of the maximum size of DNA fragment synthesis.
Finally, in order to determine the fidelity of the Minopol polymerase in the optimally functioning buffers, a series of PCR products were prepared. These products were analyzed in a Next Generation Sequencing to find the rate of error input during DNA synthesis.
