Abstract |
In this project ,the growth of the bacterium Chlorobaculum tepidum in sulfide and thiosulphate salts was studied. Chlorobaculum tepidum is a gram negative green sulfur bacterium. It makes anaerobic photosynthesis using as electron donors inorganic sulfur compounds. These compounds are mainly sulfide and thiosulfate salts which are oxidized to sulfates with the intermediate metabolite elemental sulfur originally produced and stored extracellular for later use as an electron donor when the other donors are depleted. The sulfur metabolism of the bacterium is carried to the cell periplasm and the accurate metabolic pathways are not fully known.
In this study the bacterium grown under three different conditions regards the available sulfur source. In condition 1 coexisted sulfide and thiosulfate in condition 2 thiosulfate was the sole donor and condition 3 sulfide was the sole donor. Cultures were grown under anaerobic conditions at 49 oC.
To study the growth of the bacterium we used spectrophotometric methods for determining the concentration of the bacteriochlorophyll. For studying the concentration of some bacterial metabolites we used spectrophotometric and chromatographic methods. It was observed that in condition 1 the bacterium grew faster rather than condition 3.
For further study of sulfur metabolism we invented and tested and elemental sulfur isolation protocol producing bacterium as a metabolic intermediate. Elemental sulfur was studied by electron microscopy, and mass spectrometry methods. Proteins were not found by these methods, and the elemental analysis of the samples shows that they consist mainly of sulfur which forms beads of varying size, sometimes larger than the size of the cell
Finally, it was attempted isolating the periplasmic proteome. A comparison of the three conditions proteome dual dimension IEF -SDS PAGE gel was carried out in addition with identification by mass spectrometry of proteins which were isolated from culture grown in condition 1.The identified proteins perform different functions in the cell between these proteins associated with sulfur metabolism
