Abstract The non-autonomous transposable element, Minos, was first discovered in Drosophila hydei and is a member of the superfamily Tc1/mariner transposable elements. Members of the superfamily are found in mammals, fish, nematodes and insects. Minos ability to transpose in the presence of its corresponding transposase and create stable transgenic lines in various organisms, although phylogenetically distant, made Minos a promising genetic tool for enhancer trapping, identifying and functionally analyzing new genes. Minos has already been used in a variety of model organisms such as flies, mosquitoes, sea urchins, mice and mammalian cell cultures but not in a plant model yet. In this study, we attempt to create A.thaliana Wt and dcl3- transgenic lines which carry Minos and its corresponding transposase. In this way, we aim to study Minos mobility in the plant system both in relation to the expression of the transposase and the presence of the mutated background of the RNA-dependent DNA methylation pathway (RdDM). The importance of this study is indicated as many plant species with economic and industrial interest have not been sequenced yet; moreover, many of these do not own an efficient transformation system that would enable their biotechnological study. Solution to the above biotechnological problem could give a mutagenesis system based on the controlled transfer of Minos. In the next part of this study, we examine the mRNA levels, of some of the photosynthetic apparatus genes in transgenic A.thaliana and N.benthamiana plants containing GUS and GFP transgene respectively. We study silenced and non silenced tissue concerning the expression of the transgene. The obtained results contradict previous studies as no statistical difference in gene expression in any of the conditions studied, could be observed.
