Abstract Acute lung injury (Acute Lung Injury- ALI), is being characterized by the damage of the alveolar epithelium, which results in pulmonary edema with high mortality rate. The alveolar edema develops by the leakage of protein rich fluid into the interstitium and alveolar spacies. Active ion transport is responsible for the Alveolar Fluid Clearance (AFC). Previous studies have reported that the proteins which are involved in alveolar fluid clearance are the CFTR, ENaC and the Na, K-ATPase through Cl-, Na + and Na+ '/ K + ion transport, respectively. Previous study of the expression levels of miRNAs in mice showed that miR-146 increased after induction of lung injury with ventilator. The expression levels of CFTR, NHERF1 and ENaCb genes were studied in mouse lungs after ventilator induced lung injury (VILI) with or without infusion of miR-146 and anti-miR-146. Also I tested the expression levels of NHERF1 and ENaCa genes in H1299 cells after transfection with miR-146. The data suggest that none of the genes of interest are direct targets of miR-146. Infusion with miR-146 and anti-miR-146 affect in a similar way the expression of genes. NHERF1 CFTR gene expression seem to be downregulated in miR-146/VILI and anti-miR-146/VILI mice as compared with VILI mice. Probably this downregulation occurs because of the recognition of miR-146 and anti-miR-146 by the Toll Like Receptors 7 and 8 (TLR7 / TLR8). TLR7/8 recognize single stranded RNA (ssRNA), and induce the expression of type I interferons (type I IFN) and pre-anti-inflammatory cytokines which downregulate the genes of interest.
