Abstract Tetranychus urticae and Bemicia tabaci are two major argicultural pests. Their control is largely based on insecticides. However, these pests have developed high levels of insecticides resistance. The elucidation of resistance mechanisms is an important pre-requirement for the management of resistance. In previous studies, microarray analysis of resistant T.urticae strains to the ketoenols has identified the over expression of Cyp392e10 and Cyp392e7 genes, while the Cyp392d2, Cyp392d8 and Cyp392d10 genes have overexpressed in resistant T.urticae strains to the METI's acaricides. In this study, we cloned these genes to the appropriate vectors for protein expression and we expressed the recombinant proteins CYP392E10 and CYP392E7. We have identified the capability of CYP392E10 to metabolize spirodiclofen.The P450 CYP392E10 is the first enzyme from argicultural pests shown to metabolize members of the novel ketoenol class of insecticides. In parallel, we tried to produce soluble forms of the protein CYP6CM1 (for further crystallization analysis), isolated from Bemicia tabaci that in previous studies it had been shown that this protein has the capability to metabolize the neonicotinoid imidacloprid, the number one insecticide in the world market.
