Abstract Nitrilases are enzymes that catalyze the hydrolysis of nitriles to the corresponding carboxylic acids. Studies of whole cells that exhibit nitrilase activity showed that these enzymes could act enantiospecifically when acting in prochiral and chiral nitriles. This dissertation deals with the hydrolysis of nitriles, important molecules in organic chemistry, by seven different nitrilases. Firstly, the reactivity of senen nitrilases, was studied when acting in aromatic nitriles. Most of the nitrilases showed increased reactivity hydrolyzing quantitatively nitriles much faster than their conventional chemical hydrolysis. Secondly, the enantioselectivity of seven nitrilases was studied when acting in chiral nitriles for the formation of the corresponding optically active carboxylic acids. Nitrilases in several cases showed satisfactory enantioselectivity. Thirdly, the regioselectivity of seven nitrilases was studied when acting in pro-chiral dinitriles. Unfortunately no nitrilase showed any reactivity for such substrates. Finally, one nitrilase hydrolyzed 2-methylglutaronitrile quantitatively to the corresponding monoamide in a regioselective manner.
