Abstract The goal of this project was the study of the periplasm and outer membrane proteome of the green sulfur bacterium Chlorobaculum tepidum in two different stages of growth under the prism of sulfur metabolism. The two stages were the exponential phase where the organism deploys elemental sulfur in the form of globules in the extracellular space and later on it regains it to fully oxidize it to sulfate and the at the phase was the static where the elemental sulfur was depleted In the begin, the organism was cultivated and were the growth reached the desired point then the cells were collected after centrifugation and after that the proteome isolation was conducted. The isolation of the periplasm proteome was achieved after osmotic shock and the isolation of the outer membrane proteome became possible after selective solubilization of the membrane proteome After that the proteome analysis conducted by electrophoresis of two coupled SDS gels and the identification of proteins was achieved with MALDI-TOF MS/MS
