Abstract Agrobacterium is considered as nature’s genetic engineer. Agrobacterium-mediated transient expression is used widely in many aspects of molecular biology, but the efficiency of the method depends on a variety of parameters, making the method less reliable. This study attempted to define the parameters which are involved in transient expression in N. benthamiana and use them in a more efficient-manner. Four Agrobacterium strains were studied and two of them, LBA4404 and C58C1 showed great potential. Also, the optimal optical density was determined. Additionally, evidence that support the influence of the plant and leaf age in transient expression is presented. Nevertheless, this study demonstrates the crucial role of the incubation temperature post infiltration by establishing a relation between temperature, RNA silencing and transient expression. Finally, an enhanced protocol for Agroinfiltration in N. benthamiana is proposed, taking into consideration the most important factors in order to accomplish a substantially increased efficiency.
