Abstract |
Phthalates are a group of industrial chemicals used in consumer products as solvents, additives and plasticizers. Several phthalates are known to cause carcinogenic, reproductive and development toxicities in animals. However, data on the effects of phthalate exposure in humans are still limited.
Humans can be exposed to phthalates through food, water, air and using phthalate containing consumer products. After exposure, phthalates are rapidly metabolized to their respective hydrolytic monoesters. For some phthalates, the monoesters can be further metabolized to their oxidative products before excretion in urine or feces. These metabolites have been used as biomarkers of exposure to phthalates.
The aim of the present study was the qualitative and quantitative analysis of six phthalates metabolites including monoethyl phthalate (mEP), mono-2-ethylhexyl- phthalate (mEHP), mono (2-ethyl-5-hydroxyhexyl) phthalate (mEHHP), mono (2-ethyl-5-oxohexyl) phthalate (mEOHP), mono-n-butyl-phthalate (mBP) and mono-iso-nonyl phthalate (mNP). Urine samples were collected by 81 men from various workplaces in order to assess their exposure to phthalates. Σhe analytical procedure used in this study was liquid chromatography –electrospray ionization-tandem mass spectrometry.
The highest median values of primary metabolites found in this study were found for monoethyl phthalate (mEP: 62,33 μg/g creatinine), followed by mono-n-butyl-phthalate (mBP: 20,14 μg/g creatinine) and finally mono-2-ethylhexyl- phthalate (mEHP: 10,60 μg/g creatinine). The median concentrations of the oxidized metabolites of DEHP were 27.18 μg/g creatinine for mono (2-ethyl-5-hydroxyhexyl) phthalate (mEHHP) and 15,46 μg/g creatinine for mono (2-ethyl-5-oxohexyl) phthalate (mEOHP). The primary metabolite of DiNP was not detected in any sample and therefore is not a sensitive biomarker of DiNP exposure.
Phthalate metabolite levels did not significantly differ among the groups apart from the hairdressers who are highly exposed to diethyl phthalate, DEP.
