Abstract 1 ABSTRACT Many studies have shown that HDL has a plethora of functions that may contribute to the protection from cardiovascular disease. The pathway of the biogenesis and catabolism of HDL is a complex process and involves several membrane bound and plasma proteins. Using adenoviruses mediated gene transfer in various mouse models, the studies described in this thesis explore the pathway of the biogenesis of HDL and more specifically the following: 1. The importance of the hydrophobic residues in the 218-230 region of apoA-I in the biogenesis of HDL. 2. The effects of LCAT mutations on the biogenesis of HDL. 3. The effects of CTα on the biogenesis of HDL. 4. The contribution of dominant mutations in apoE to lipid homeostasis and the biogenesis of HDL. 5. The synergy of apoA-IV and LCAT on the biogenesis of apoA-IV containing HDL. 6. The effect of reconsituted HDL containing apoE and apoA-I on endothelial gene expression.
