Abstract In the present work, the effect of growth conditions on the production of exopolysaccharides by photosynthetic microorganisms was investigated. The experiment was divided in two parts: In the first one, the microorganism was cultivated without air inflow under stress conditions for the cells, which was achieved by removing nitrogen from the growth medium. Also, the effect of a carbon source, like sodium carbonate, carbon dioxide, glucose and sodium acetate, on the production of exopolysaccharides by the cyanobacterium Nostoc calcicola was investigated, due to the absence of any other carbon source in the medium. In the second part of the experiment, EPS production with air inflow was studied in four different growth conditions, in the presence of all components, in the absence of nitrogen, in the absence of both nitrogen and carbon, and in the absence of sulfur. Finally, in these conditions, the cells were studied by optical microscopy. In the aforementioned cultivations, the cell growth (concentration of chlorophyll a and dry weight), the concentration of exopolysaccharides and the quantity of peptides in the supernatant fluid of the cultures were measured at regular intervals.
