Abstract Telomeres are structures of DNA and proteins that form complexes that protect the ends of chromosomes in all organisms. Understanding the conservation mechanisms of telomeres and the ways in which their function is affected has been significantly studied in recent years. During our lifetime, the length of telomere ends in chromosomal DNA molecules decreases as a result of various factors. The aim of the present study was to qualitatively and quantitatively determine the length of chromosomal DNA telomeres, to investigate the effect of various endocrine disruptors on them. More specifically, the disruptors studied were Parabens (PBs), Triclosan (TCS), Bisphenol A (BPA) and Glyphosate. In this study, the effect of the aforementioned endocrine disruptors on chromosomal DNA telomere length in the rabbit thyroid gland was investigated. The study group consisted of four groups of four rabbits (2 males, 2 females) of which each group was given, for the same period (12 consecutive months), a different amount and combination of endocrine disruptors. More specifically, the first study group was the control group as, in its members were not administered endocrine disruptors, while the other three groups were administered one or a mixture of disruptors in high or low doses. More specifically, Group 2: Low dose BPA / PBs / TCS / glyphosate, Group 3: High dose BPA / PBs / TCS / glyphosate and Group 4: High dose glyphosate - commercial form). The respective dose was administered once a day, five days a week. At the end of the year, the rabbits were hypnotized and killed in order to receive part of their thyroid gland. To determine the effect of endocrine disruptors on telomere length, their length was measured by RTqPCR where the results were obtained in the form of fluorescence intensity diagrams as a function of the numbers of polymerization reaction cycles. The quantification of the reduction in length was performed using the average length of the telomeres of each group determined experimentally, with the average of the control group having the role of the reference length. Quantification results showed a decrease in telomere lengths in rabbit thyroid tissue. This reduction was evident both in members receiving only a single substance and in those receiving a mixture of substances, indicating that EDs have genotoxic effects on DNA and in particular on endocrine tissue DNA. The genotoxic effect on the DNA of the thyroid gland and specifically on telomere lengths is evidence of the effect that various EDs have on cell aging and cell apoptosis. These results indicate the need for further study regarding the genotoxic effect of EDs on the endocrine system in both experimental animals and humans.
