Abstract Melanoma is the deadliest skin malignancy, and its incidence is constantly increasing. The diversity in the biological behavior of the disease, makes it a difficult therapeutic and sometimes a diagnostic goal. Early diagnosis is important for prognosis, disease progression and treatment options Microphthalmia-associated Transcription Factor (MITF) is a transcription factor that plays a central role in both normal melanocyte biology and melanoma biology, as it controls key signaling pathways. MITF expression levels as well as their combination with the expression levels of other transcription factors (eg FN-1, AXL, D2-40), can provide valuable information on disease progression, prognosis and response to treatment. Thus, MITF is used as a diagnostic immunohistochemical biomarker, as a molecular biomarker of metastasis by PCR analysis, as genetic screening for disease predisposition, is included in clinical diagnosis codes while at the same time the mutations of its gene are actively investigated. Precisely because MITF expression levels act as a "rheostat" for disease progression, research has been conducted for the past 20 years to discover substances that either directly affect MITF or affect its expression levels. The resulting substances are of plant origin, are products of chemical processes, are products of biological processes of other organisms (eg viruses, fungi) or finally are drugs that are used for various diseases (eg blood sugar control) but can, through appropriate pathways, influence MITF levels. MITF is essential in understanding the biology of melanoma and in managing the disease. Its role as a biomarker is particularly important as it helps in the timely and accurate diagnosis of the disease. As a goal of treatment, it gives hope for the final treatment of melanoma in the future.
