Abstract The synthesis of Mn2+ ion-selective markers has been a long time challenge for the research community, given the difficulty of the construction of cation chelator systems that would be selective for the specific target ion. In the context of the present study, the synthesis of a small series of fluorescent markersderivatives of 7-dimethylaminocoumarin was performed, followed by the study of their fluorescence profile. It was decided that (a) the fluorophore would contain the chromophore group of coumarin, while (b) the ionophore would alternatively contain either a carboxyl group, therefore the probe would contain two carboxyl units (one in the form of the coumarin lactone) or two pyridine groups, in an attempt to imitate the dipyridine system of the Liang and Canary probes which were the only indicators that were reported as selective for Mn2+ ions The markers that were synthesized and studied are (a) 7-(dimethylamino)coumarin (1d), (b) 4-carboxy-7-(dimethylamino)coumarin (1bc-ONα) and (c) 7-(dimethylamino)-2-oxo-N,Nbis(pyridin-2-ylmethyl)-2H-chromo-3-carboxamide (4c) All indicators exhibited a "turn-off" response, meaning that the increase of ion concentration leads to a decrease in the relative fluorescence intensity. Kd values are in the micromolar (mM) range. Fluorescence profile studies showed that the presence of the di-(2-picolyl)amine group contributes to the selectivity for Mn2+ ions, which was the main goal of the research project. Speciation studies indicate that the 4c / Mn2+ ratio is most likely 2:1, i.e. the system behaves as a tetradentate chelator complex. Ion competition studies have shown that acetonitrile solutions have a distinct selectivity for Mn2+ ions. In conclusion, although fluorescence indicator 4c cannot function as a Mn2+ indicator in biological (aqueous) systems, it can act as an analytical tool for the determination of Mn2+ levels in acetonitrile analytical samples.
