Abstract The hEMSY consists of 1322 amino acids and has been shown to take part in familiar breast cancer cases through interaction with BRCA2, as well as in same sporadic ovarian and breast cancer cases, where its gene is multiplied. It contains an 80-amino acid sequence in the EMSY N-Terminus, thus called ENT, which has proven to be conserved among several species. EMSY interacts with chromatin regulators and forms homodimers through binding of the ΕΝΤ domains. In Homo sapiens, there is only one copy of the ENT gene, while in Arabidopsis thaliana, 9 proteins have been discovered to contain the ENT domain˙ 8 of them also include a Tudor-like domain, called Agenet. These EMSY-like (EML) proteins in plants, communicate with chromatin remodelling factors and other molecules that participate in the epigenetic gene regulation in the process of flowering. The aim of our project was to biochemically characterise the Agenet and ENT domains. After heterologous expression in bacterial E. coli cells, we tried to confirm the DNA binding capacity of the ENT domain, as well as shed light into the binding specificity of the Agenet domain for different modifications in histone tail residues.
