Abstract Selenium constitutes a nutritious trace element for the human organism, which is taken up via food. Concretely the more important sources of selenium are fish, meat, liver, nuts and cereals. Thus in the present study was developed a method for the total determination of selenium with Mass Spectrometry Inductively Coupled Plasma. The results from the total determination of selenium in the various foods, are combined with DAFNE (Data Food Networking), which aims in the diachronic follow-up and comparison of alimentary habits of European populations and concretely the Greeks and gives us information on the medium consumption of a Greek, in grams per day, so that we can calculate the quantitative intake of Se for a Greek.
Aiming at the speciation of selenium species were developed various methods of extraction (hydrolysis extraction, extraction with buffer, enzymatic extraction), in two kinds of fish, for the suitable method that will give the better desirable results and concrete, the higher recovery of the extraction. The higher recovery of the extraction has been found when we used the enzymatic digestion with the addition of enzyme Pronase E.
Also an in vitro gastrointestinal method was accomplished for the determination of % bioavailability of selenium, the quantity of selenium which is absorbed by the human organism, when he consumed the two kinds of fish. In vitro gastrointestinal digestion includes a simulated gastrointestinal digestion with the pepsin in gastric stage (juice) and a mixture of pancreatin, bile salts and a-amylase at the intestinal stage (juice).
The extracts of the enzymatic and gastrointestinal digestion have been analyzed with HPLC-ICP-MS (reversed phase chromatography). The results of speciation analysis proved that the dominant form of selenium in the two kinds of fish is selenomethionine (SeMet).
