Abstract This M.Sc. thesis focuses on the compositional analysis of milks of different animal origin, as well as different milking period using MMR spectroscopy and chemometrics. Towards this purpose, a large number of milk samples obtained from different animal species and originated from commercial farms in northern Greece, were studied. Specifically, the milk samples were collected from cow, sheep, goat, buffalo and donkey species with the different milk production period ranging between five (March to July for cows) and two months (June to July for donkeys). The experimental protocol used for milk analysis involved the extraction of polar (sugars, low molecular weight organic acids and amino acids) and non-polar metabolites (fat such as triglycerides, diglycerides and linolenic) using two different solvents. This was followed by, the successful identification of milk metabolites present in these two extracts, as well as their quantification based on their NMR spectra. The analytical compositional data set obtained for the 91 samples analyzed was used to examine milk composition variations according to animal origin and milking period. Following that, the techniques of targeted (metabolite profile) and untargeted (bucketed 1H and 31P NMR spectra) metabolomics were applied to the milk samples datasets obtained using suitable multivariate statistical analysis methods. The metabolomics analysis led to the development of various models that can successfully differentiate milk samples according to their animal origin (cow, sheep, goat, buffalo and donkey) and the period of milk collection, while the metabolites that are statistically significant for the discrimination of the different milk categories were also identified.
