ενεργοποίηση της RhoA από τον TGF-β. Επίσης δείχθηκε, ότι η Net1A αποτελεί άμεσο
μεταγραφικό στόχο του TGF-β και ότι η επιλεκτική έκφραση της έναντι της Net1 δεν οφείλεται
στην ενεργοποίηση κάποιου ειδικού υποκινητή, αλλά πιθανότατα στη ρύθμιση του
εναλλακτικού ματίσματος από τον TGF-β. Το Smad μονοπάτι – κυρίως η Smad3 – καθώς και
το ERK MAPK μονοπάτι δείχθηκε ότι εμπλέκονται στο μηχανισμό έκφρασης της Net1A από
τον TGF-β
Η μακρόχρονη επίδραση με TGF-β μείωσε τα επίπεδα mRNA της Net1 ισομορφής και
οδήγησε τη Net1A για αποικοδόμηση από το πρωτεόσωμα. Επιπλέον, αναγνωρίστηκαν τα
microRNAs, miR-24 και miR-155, ως νέοι μετα-μεταγραφικοί ρυθμιστές της έκφρασης της
Net1A, τα οποία επάγονται από τον TGF-β και στοχεύουν άμεσα την 3’UTR του mRNA της
Net1A (και της Net1). H αποσιώπηση και μόνο της Net1A απορύθμισε τις διακυτταρικές
συνδέσεις που μεσολαβούνται από την E-cadherin και τη ΖΟ-1 και οδήγησε στην έκφραση
των μεσεγχυματικών δεικτών N-cadherin, fibronectin, PAI-1 και του καταστολέα της Ecadherin,
Slug, ενώ ενίσχυσε το μεσεγχυματικό φαινότυπο μετά από επίδραση με TGF-β,
υποδηλώνοντας ότι η μείωση των επιπέδων της Net1A μετά από μακρόχρονη επίδραση με
TGF-β παίζει ρόλο στην επιθηλιομεσεγχυματική μετατροπή (EMT). Το miR-24 δείχθηκε επίσης
ότι εμπλέκεται στο μηχανισμό της επαγόμενης από τον TGF-β ΕΜΤ και ότι παίζει σημαντικό
ρόλο στη διεισδυτικότητα καρκινικών κυττάρων μαστού που ρυθμίζεται από τον TGF-β μέσω
στόχευσης στη Net1A. Τέλος, μετά από ανάλυση 6 ζευγών πρωτογενούς και μεταστατικού
καρκίνου του μαστού, τα επίπεδα του miR-24 βρέθηκαν αυξημένα, ενώ τα επίπεδα της Net1A
ελαττωμένα στους μεταστατικούς σε σχέση με τους πρωτογενείς όγκους, υποδηλώνοντας ότι
το μονοπάτι miR-24/Net1A ενεργοποιείται κατά τη μεταστατική διαδικασία στον καρκίνο του
μαστού
Συμπερασματικά, στην παρούσα διατριβή παρουσιάζεται ένας νέος μηχανισμός που
ρυθμίζει τη γρήγορη και παρατεταμένη αναδιοργάνωση της ακτίνης μέσω των Rho GTPασών
και ελέγχει διάφορες κυτταρικές διεργασίες, όπως η οργάνωση του κυτταροσκελετού, η ΕΜΤ
και η διεισδυτικότητα τόσο σε φυσιολογικά, όσο και σε καρκινικά κύτταρα.

Summary
Transforming growth factor-β (TGF-β) controls many fundamental aspects of cellular behavior,
such as cell division, differentiation and migration, which require active reorganization of actin
and microtubule cytoskeleton and regulation of contractile forces.
TGF-β has been shown to induce short- and long-term actin reorganization by
controlling Rho-GTPase signaling in a variety of cell types, such as fibroblasts, epithelial and
smooth muscle cells. A number of direct target genes, activated by TGF-β, have been
previously reported to control the long-term Rho activation and actin reorganization, such as
α-smooth muscle actin (α-SMA), RhoB GTPase and the guanine nucleotide exchange factor
(GEF) Net1. However, the molecular mechanisms that regulate the prompt and long term
stimulation of Rho GTPases by TGF-β remain poorly understood. The purpose of the present
study was to delineate the mechanisms of the rapid and long term activation of Rho GTPase
by TGF-β, as well as to study their role in actin cytoskeleton reorganization and in biological
processes such as epithelial to mesenchymal transition (EMT) and cancer cell invasiveness.
By using JEG3, choriocarcinoma cells that lack endogenous Smad3 we found that TGF-
β rapidly stimulated RhoA and RhoB activation, whereas inhibition of Smad2 expression via
siRNA-mediated silencing or by blocking its phosphorylation using the TβRI inhibitor,
SB431542, did not prevent the early RhoA/B activation by TGF-β, indicating that this effect is
Smad2/3-independent. When we pre-treated cells with the general tyrosine kinase inhibitor
Genistein we observed that the TGF-β-induced early RhoA activation was blocked, implying
that tyrosine phosphorylation signaling is involved in this process. In line with this finding,
TGF-β-stimulation resulted in a quick activation of the non-receptor tyrosine kinase Src,
followed by activation of the Vav2 GEF. Furthermore, the early TGF-β-induced RhoA
activation was totally blocked after inhibition of Src kinase by the selective inhibitor of the Src
family tyrosine kinases PP2 and was reduced after Vav2 silencing via siRNA, supporting the
notion that the rapid Src/Vav2 stimulation is implicated in the regulation of early RhoA
activation by TGF-β.
In parallel, we studied the mechanism of long term activation of RhoA GTPase by TGF-β
by focusing on the regulation of the two isoforms of the RhoA-specific Net1 GEF by TGF-β in
HaCaT keratinocytes. We found that short-term TGF-β treatment selectively induced Net1
isoform2 (Net1A) but not Net1 isoform1. This led to up-regulation of cytoplasmic Net1A protein
levels that were necessary for TGF-β-mediated RhoA activation. We also showed that Net1A
is a direct transcriptional target of TGF-β and its selective induction is not due to the activation
of a unique promoter, but rather due to regulation of alternative splicing by TGF-β. Smad
signaling – mainly Smad3 – and the ERK MAPK pathway were found to be involved in Net1A
up-regulation by TGF-β.

Interestingly, long-term TGF-β treatment resulted in Net1 mRNA down-regulation and
Net1A protein degradation by the proteasome. Furthermore, we identified the microRNAs,
miR-24 and miR-155, as novel post-transcriptional regulators of Net1A expression, which are
upregulated by TGF-β and directly target the 3’UTR of Net1A (and Net1) mRNA. Silencing of
Net1A alone resulted in disruption of E-cadherin and zonula occludens-1 (ZO-1) mediated
junctions, as well as expression of the mesenchymal markers N-cadherin, fibronectin and PAI-
1 and the transcriptional repressor of E-cadherin, Slug and enhanced the mesenchymal
phenotype in the presence of TGF-β stimulation, indicating that late TGF-β-induced
downregulation of Net1A is involved in epithelial to mesenchymal transition (EMT). The
microRNA miR-24 was also found to be implicated in the regulation of the EMT program in
response to TGF-β and was shown to be directly involved in the TGF-β induced breast cancer
cell invasiveness through Net1A regulation. Finally, after analyzing 6 pairs of primary and
metastatic breast tumors we found that miR-24 was upregulated and Net1A downregulated in
metastatic versus primary tumors, implying that the miR-24/Net1A pathway is activated during
breast cancer metastasis.
In conclusion, the present study presents a novel mechanism which regulates the shortand
long-term actin reorganization through Rho GTPase signaling and controls cellular
responses such as cytoskeleton organization, EMT and invasiveness in normal and tumor
cells.
