Abstract Mitochondria are significant organelles of the eukaryotic cells that contain two membranes, the outer (OM) and the inner (IM) membrane, enclosing two compartments, the matrix and the inner membrane space (IMS). Proteins of relatively small molecular mass, localized in the IMS, often contain conserved cysteines arranged in CXnC motifs [53]. Such proteins perform important functions in the cell, including assembly of the respiratory chain and mitochondrial protein biogenesis. The import of these cysteine-containing proteins is mediated by a disulfide relay system in the IMS, known as the Mitochondrial Intermembrane space Assembly (MIA) machinery. Components of this specific system that traps precursor proteins in mitochondria are the oxidoreductase Mia40 and the sulfhydryl oxidase Erv1 [11, 54]. Mia40 is the key protein that introduces disulfide bonds in the substrates during passage to the IMS, thereby locking them in a folded conformation (oxidative folding). It contains six conserved cysteines that form the active site CPC and a duplicated structural CX9C motif. Erv1 on the other hand, is a flavoprotein that re-oxidizes Mia40 to its active state. In this study, the main goal is to investigate the molecular mechanism and determinants of biogenesis of Mia40. We attempt to dissect the import stages of Mia40 into the IMS. Experiments include in vitro import assays with isolated yeast mitochondria, in vivo assays with yeast cells from the strain GALMia40 and ITC binding experiments with isolated proteins.
