Abstract |
The study of proteins and protein complexes of bacterial origin is an area of growing
interest in the research of transporting and metabolizing the nutrients of a bacterium.
Pseudomonas sp. phDV1, is a Gram-negative bacterium selected for its ability to
degrade aromatic compounds such as phenol and ortho-cresol. In the present study, the
bacterium was cultured using succinic acid, phenol, and ortho-cresol as sole carbon
sources, respectively, for the purpose of proteomic analysis of water-permeable and
membrane proteins / complexes of the bacterium.
In order to achieve the growth of the bacterium, guided by the toxicity of the above
aromatic compounds, culture attempts were made until the growth of the bacterium in
the respective pollutant was achieved. The biological samples were collected from
which the cytoplasmic and membrane fraction were isolated. This is followed by
analysis of the protein fractions with Blue Native PAGE (1D and / or 2D
electrophoresis). The identification of the separated proteins was achieved by mass
spectrometry and more specifically by using Matrix Assisted Laser Desorption
Ionization (MALDI -TOF MS / MS) and nano-LC-Electron Spray (ESI) MS / MS. The
results obtained from the proteomic analysis are comparative studies of proteins and
protein complexes that are identified according to each condition.
The present work is a new approach to the study of Pseudomonas sp. phDV1, in the
presence of monocyclic aromatic pollutants.
